I have an urethane super natural with a layout 30°x 3 3/4 x 30° and is OK for one or two games then start hooking too much and needs to start open the lines and don't really like that with an urethane bowling ball. I'll prefer to play straight.
I have another super natural ball undrilled, but wants to make it longer and strong back end, what kind of layout would you guys recommend ???????

Your existing 30°x 3 3/4 x 30° is approximately 70-3 3/4-30 without a low balance hole.
Option one: Long and strong as compared to existing, utilize a Motion Hole with an 80-4-30 layout.
Option 2 is a weaker overall Control layout to your existing, low flare 70-5-35
Layouts do not work miracles.
You are very rev dominant and the lanes are becoming dry enough to make the ball you are rolling coverstock dominant and you refuse to move left into the oil.
Utilizing surface between the 2 will help keep the reaction from being too close to each other. Playing your existing one dull, 2000 grit, and the new one shiny, 4000+ will help.
If you continue to fight the lanes I suggest rolling plastic to stay in your comfort zone of playing straight.

Thanks Erick for you response, I will try as you said resurface both ball and also going to try the 1st layout to see how it works for me, Most of the time always have to play deep inside, but now I try something different that's why I come up with urethane.

More and more bowlers are using urethane balls. Does the same sweet spot calculation apply still? Any extra factors to be aware of when laying out urethane balls?

The Sweetspot calculations are about the core.
Layouts, from there, can be adjusted to accommodate different coverstock characteristics.
I still don't believe there is a need to adjust the Sweetspot calculations.
